Design Notes
Due to the small size and lack of side chain bulk in glycine, the GGGS linker provides an ideal degree of flexibility. This flexibility allows the linked protein domains to move independently, ensuring that each domain can fold correctly and maintain its functional conformation in Saccharomyces cerevisiae. The serine residues add hydrophilicity to the linker, improving solubility and reducing the risk of aggregation in the fusion protein.
